Dynamics of Golgi resident proteins in the newly forming Golgi units. ManII-HRP-expressing HeLa cells were incubated with DAB and H2O2 to cross-link the preexisting (old) Golgi and fixed for IF either immediately or after incubation with fresh medium for 2, 6, 10, or 24 h. DAB-treated and non-treated (NT) cells were stained for GM130 (green) and another Golgi marker (red), as indicated.

Cargoes move vectorially from cis- to trans-Golgi. (A) HeLa cells were transfected with VSVG-GFP-RUSH constructs overnight and then treated with nocodazole (33 uM) for 3 h at 37 degrees C. Biotin was then added to cells and at indicated times the cells were fixed and stained for GM130 and TGN46 to mark the cis- and trans-Golgi, respectively. The relative position of the cargo with respect to GM130 and TGN46 was measured.

Representative images of HeLa-K cells stained with antibodies against DDR proteins and the Golgi marker, GM130. Cells were treated with a control or an siRNA against the protein of interest to validate the specificity of the antibody. The changes in intensity measurements in the Golgi and nuclear compartment were quantified (n = 2 for all proteins except for POLQ, which had n = 1, biologically independent experiments with at least 200 cells analyzed for each treatment). Scale bar, 10 μm. (human (HeLa-K cells); Figure S1, full figure.)
