Osteocyte LCN (OLCN) revealed by acid-etched, resin-casted SEM imaging. Representative acid-etched SEM images showing the structure of osteocyte LCN in the iliac bone tissues from an 11.4-yr-old female (control). A, B) Low-magnification (×400) SEM images. C, D) Medium-magnification (×1000) SEM images. E, F) High-magnification (×2000) SEM images. Control panels (left column, A, C, E) show normal lacunar shape and canalicular length and number. (human (iliac bone biopsy, non-AIS control, female aged 11.4 yr); Figure 1, control panels (A, C, E).)

Peri-osteocytic lesions (POLs) and osteocyte cell-matrix/mineral relationships. (A) Contact microradiograph of a human bone slice showing dark, unmineralized peri-osteocytic lesions (halos) surrounding osteocyte lacunae in osteomalacic bone from a patient with X-linked hypophosphataemia (XLH). (human (iliac crest bone biopsy, X-linked hypophosphataemia); Figure 5, A.)

Histological section from the parallel lamellar bone in ROI 3, showing osteocyte lacunae and canaliculi in Giemsa staining. Panel (A) shows individual osteocyte lacunae with their canalicular processes extending through the lamellar matrix. Panel (B) shows the osteocyte lacuno-canalicular network (OLCN) visualized by Giemsa staining in a bone sample from a patient with pycnodysostosis, with well-preserved osteocyte morphology in the parallel lamellar region. (Homo sapiens; Figure 4.)

Goldner stained section of cortical bone (ROI 3) viewed under polarized light. Two distinct regions are observed: (A) a shell of primary (non-remodeled) bone formed by periosteal apposition, consisting of parallel lamellae along the periosteum, and (B) intra-cortically remodeled osteonal bone with concentrically arranged lamellae around a central Haversian canal. Osteocyte lacunae are distributed in the lamellar matrix in both regions. Human surgical bone sample from a patient with pycnodysostosis (cathepsin K deficiency). (Homo sapiens; Figure 3.)

Osteocyte network characteristics in human bone biopsies. Representative toluidine blue stained histological images of trabecular bone. Viable osteocyte lacunae with cell nuclei are evident in the bone matrix. Both treatment-naive patients and those after denosumab therapy are shown, demonstrating the osteocyte lacuno-canalicular network in trabecular human iliac crest bone. (Homo sapiens; Figure 4.)
